I'd say NetHack, easily. The game has tons of variables and randomization, you learn something new with each game. And it has been stated to take years to master (though I somehow got my first ascension in about 3-months play.) If I ever got stuck on a desert island, and could only have one game to spend the rest of eternity with, NetHack would be it. Enough said.
Close favorites would be DooM/DooM II/Final DooM/Ultimate DooM, plus all WADs/TCs ever created for them, all conveniently loaded up on Doomsday. Those would keep me busy for quite a while.
